建立大鼠矽肺模型及对照,分别取不同时间点处死大鼠,免疫组化检测肺组织中uPA、PAI-1蛋白表达。与对照组比较,矽肺模型组uPA表达随时间延长呈增强趋势,第1周达高峰,此后逐渐减弱,但仍较对照组明显。PAI-1第1天起即可见阳性表达,且随染尘时间延长强度逐渐增强,持续至第4周。模型组肺内多种细胞uPA、PAI-1表达增加,矽肺纤维化中可能存在uPA/PAI-1比例的失衡,导致细胞外基质(ECM)积聚而影响矽肺纤维化的发生发展。
Rat silicosis model and control were established. The rats were sacrificed at different time points, and the expressions of uPA and PAI-1 protein in lung tissues were detected by immunohistochemistry. Compared with the control group, the expression of uPA in silicosis model group increased with time and peaked in the first week, then gradually decreased, but still significantly higher than that in the control group. The positive expression of PAI-1 was observed on the first day, and gradually increased with the prolongation of the dust exposure, lasting for the fourth week. The expressions of uPA and PAI-1 in many kinds of cells in the model group increased. The imbalance of uPA / PAI-1 ratio in silicotic fibrosis may lead to the accumulation of extracellular matrix (ECM), which may affect the occurrence and development of silicosis fibrosis.
