采用紫外分光光度法测定明胶微球中甲氨蝶呤的含量。当波长为 30 3nm时 ,甲氨蝶呤浓度在 3~ 18mg/L范围内其吸收度与浓度呈良好线性关系 ,回归方程为A =0 0 5 2C - 0 0 10 (r =0 9998,n =6 )。测定两种不同批号微球的药物含量 ,平均回收率分别为 99 0 8%和 10 1 32 % ,相对标准偏差RSD分别为 0 40 %和 0 2 6 % ,总平均回收率为 10 0 2 % (n =3,RSD =0 48% )。该方法简便、快速、准确 ,并具有良好的重现性。
Determination of methotrexate in gelatin microspheres by ultraviolet spectrophotometry. When the wavelength of 30 3nm, the concentration of methotrexate in the range of 3 ~ 18mg / L showed a good linear relationship between the absorption and concentration, the regression equation was A = 0 0 5 2C - 0 0 10 (r = 0 9998, n = 6). The drug contents of two different batches of microspheres were determined. The average recoveries were 99.0% and 10.1%, respectively. The relative standard deviations (RSDs) were 0 40% and 0 26%, respectively. The average recovery was 100.02% (n = 3, RSD = 0 48%). The method is simple, fast, accurate and has good reproducibility.
