目的:研究26-nor-8-羟基-α-芒柄花醇(26-nor-8-hydroxy-α-onocerin,26HO)对成骨细胞活性的影响。方法:采用MTT法研究不同浓度的26HO对成骨细胞增殖率的影响;并用Real-Time PCR研究不同浓度的26HO对成骨细胞成骨相关基因表达量的影响。结果:20μM/L和40μM/L的26HO短期处理(1d)能够促进成骨细胞增殖,长期处理对成骨细胞增殖活性没有显著影响;26HO对骨相关基因(ALP、OCN、Col-I)表达的影响与成骨细胞的成熟状态有关。而对AP-1转录因子家族基因中Fos亚家族成员(c-fos、Fra-1、Fra-2)等基因mRNA表达的影响与26HO浓度有关,与处理时间无关;而对AP-1转录因子家族基因中Jun亚家族成员(c-Jun和Jun-D)基因mRNA表达的影响与26HO浓度无关,与处理时间有关。结论:26HO具有成骨活性。
Objective: To investigate the effect of 26-nor-8-hydroxy-α-onocerin (26HO) on osteoblast activity. Methods: MTT assay was used to study the effects of different concentrations of 26HO on the proliferation of osteoblasts. Real-time PCR was used to study the effects of different concentrations of 26HO on osteoblast-related gene expression. Results: 26HO short-term treatment (1d) of 20μM / L and 40μM / L could promote the proliferation of osteoblasts. Long-term treatment had no significant effect on the proliferation activity of osteoblasts. 26HO expressed the expression of bone related genes (ALP, OCN, Col-I) The effect is related to the maturation status of osteoblasts. The effect of AP-1 transcription factor family genes on the mRNA expression of Fos subfamily members (c-fos, Fra-1, Fra-2) and 26HO concentration had nothing to do with the treatment time; The effects of Jun-family members (c-Jun and Jun-D) mRNA expression in family genes were independent of 26HO concentration, and were related to the treatment time. Conclusion: 26HO has osteogenic activity.
