目的 :探讨细胞凋亡在宫颈癌发生发展中的变化。方法 :采用脱氧核苷酸转移酶介导的生物素末端缺口标记法 (TdTmediateddUTP biotinnickendlabeling ,TUNEL)检测 15例正常宫颈上皮 ,2 2例宫颈上皮内瘤变 (CIN)Ⅰ /Ⅱ ,16例CINⅢ及 5 2例浸润性宫颈鳞癌 (ISCC)组织中的细胞凋亡标记指数 (A LI) ,并用免疫组化ABC法检测这些组织中增殖细胞核抗原PCNA的表达。结果 :在正常宫颈上皮 ,CINⅠ /Ⅱ ,CINⅢ及ISCC中A LI分别为 0 0 0±0 0 0 ,0 33± 0 0 9,0 6 9± 0 14和 0 0 7± 0 0 1;PCNA标记指数 (P LI)分别为 :9 87± 1 32 ,2 2 5 7± 2 0 5 ,31 0 1± 2 0 4和6 5 92± 3 0 1。结论 :细胞凋亡在宫颈癌恶变前的发生发展中以抑制细胞过度增殖而发挥负调控作用 ,在ISCC阶段其调控作用可能已经紊乱。
Objective : To explore the changes of apoptosis in cervical carcinogenesis. Methods: 15 cases of normal cervical epithelium, 22 cases of cervical intraepithelial neoplasia (CIN) I/II, and 16 cases of CIN III were detected by deoxynucleotidyl transferase mediated TNT-mediated biotinnick end labeling (TUNEL). The apoptosis index (A LI) in 52 invasive cervical squamous cell carcinoma (ISCC) tissues was detected. The expression of proliferating cell nuclear antigen (PCNA) in these tissues was detected by immunohistochemical ABC method. RESULTS: In the normal cervical epithelium, A LI in CINI/II, CINIII, and ISCC were 0 0 0 ± 0 0 0, 0 33 ± 0 0 9, 0 6 9 ± 0 14 and 0 0 7 ± 0 0 1 respectively; PCNA The PLIs were: 9 87 ± 1 32, 2 25 7 ± 2 0 5, 31 0 1 ± 2 0 4 and 6 5 92 ± 3 0 1. Conclusion : Apoptosis plays a negative regulatory role in inhibiting cell hyperproliferation before and after malignant transformation of cervical cancer. Its regulation may be disorganized at the ISCC stage.
