目的:探讨中药苏木提取物诱导人类慢性髓性白血病K562细胞株的凋亡作用。方法:采用MTT法检测苏木提取物对K562细胞增殖抑制作用;荧光显微镜观察细胞形态变化;库尔特全自动颗粒粒度分析药物引起K562细胞体积大小的分布变化;琼脂糖凝胶电泳测定DNAladder及流式细胞术检测细胞周期变化。结果:25μg/ml苏木浸膏能明显诱导K562细胞凋亡,产生凋亡细胞所具有的典型形态学及生化学特征,同时对K562细胞的增殖抑制和诱导凋亡作用呈一定的浓度依赖性。结论:中药苏木提取物能诱导细胞凋亡,抑制癌细胞增殖呈一定的浓度依赖关系。
Objective: To investigate the apoptosis of human chronic myelogenous leukemia K562 cell line induced by the extract of Hemu. METHODS: MTT assay was used to detect the inhibitory effect of Hemu extract on the proliferation of K562 cells; fluorescence microscopy was used to observe the changes of cell morphology; Kurt automatic particle size analysis was used to analyze the distribution of K562 cells; Agarose gel electrophoresis was used to determine DNA ladders and Flow cytometry was used to detect changes in cell cycle. Results: 25μg/ml Hemu extract can obviously induce apoptosis of K562 cells and produce the typical morphological and biochemical characteristics of apoptotic cells. It also has a concentration-dependent effect on the proliferation inhibition and apoptosis induction of K562 cells. . Conclusion: The extract of Hemu can induce apoptosis and inhibit the proliferation of cancer cells in a concentration-dependent manner.
