本文研究了促性腺激素(FSH和LH)和促性腺激素释放激素(GnRH)类似物(GnRHa)对大鼠颗粒细胞(GC)tPA和PAI-1表达的调节。(1)FSH和LH及GnRHa或大蓟二萜醇-12-14烷基-13乙酸盐(PMA)可单独刺激GC分泌tPA;FSH(或LH)与GnRHa(或PMA)共同作用于GC时所增加的tPA活性比两者单独作用之和要高;(2)FSH和LH降低GC的PAI-1活性,而GnRHa和PMA则明显刺激GC的PAI-1活性和PAI-1 mRNA水平;(3)GnRHa和PMA刺激GC的PAI-1活性和PAI-1的mRNA水平;但在FSH或LH存在的情况下这种刺激受到完全抑制。因此促性腺激素和GnRHa对GC tPA和PAI-1表达的调控机制可能是不同的。
This article investigates the regulation of tPA and PAI-1 expression in rat granulosa cells (GCs) by gonadotropin (FSH and LH) and GnRH analogs (GnRHa). (1) FSH and LH and GnRHa or diterpene-12-14 alkyl-13 acetate (PMA) can stimulate GC secretion of tPA alone; FSH (or LH) and GnRHa (or PMA) co-act on GC (2) FSH and LH decreased the PAI-1 activity of GC, while GnRHa and PMA significantly stimulated PAI-1 activity and PAI-1 mRNA level of GC; (3) GnRHa and PMA stimulated PAI-1 activity and PAI-1 mRNA level in GC; however, this stimulation was completely inhibited in the presence of FSH or LH. Therefore, the regulatory mechanisms of gonadotropin and GnRHa on GC tPA and PAI-1 expression may be different.
