目的研究藻蓝蛋白对2型糖尿病大鼠肝脏和胰岛形态结构和细胞凋亡的影响。方法健康雄性Wistar大鼠32只,高糖高脂饲料喂养联合小剂量链脲佐菌素(STZ)腹腔注射建立2型糖尿病大鼠模型,藻蓝蛋白灌胃治疗,组织病理学技术观察肝脏和胰岛细胞的形态结构,用原位末端标记法(Tunel法)检测肝细胞和胰岛细胞凋亡。结果模型组大鼠肝组织结构紊乱,中央静脉扩张,肝细胞肿胀,颗粒变性和脂肪变性;胰岛萎缩、结构模糊,细胞数目减少、排列紊乱,胞浆肿胀、空泡变性等;两个组织中凋亡阳性细胞较正常组明显增多(P<0.05);藻蓝蛋白组肝脏组织和胰岛细胞形态结构较清晰,正常细胞数目增多、排列较规则,肿胀和变性细胞较少,凋亡阳性细胞较模型组显著减少(P<0.05)。结论藻蓝蛋白可抑制糖尿病大鼠肝脏和胰岛细胞凋亡,发挥一定的保护作用。
Objective To study the effects of phycocyanin on the morphology and apoptosis of liver and pancreatic islets in type 2 diabetic rats. Methods Thirty-two male Wistar rats were randomly divided into high glucose and high fat diet fed with low dose streptozotocin (STZ) to establish type 2 diabetic rat model. The mice were given gavage with phycocyanin and the histopathology was used to observe the liver and The morphological structure of islet cells was examined by TUNEL method. The apoptosis of hepatocytes and islet cells was detected by Tunel method. Results In the model group, the hepatic tissue structure was disorganized, the central venous dilatation, hepatocyte swelling, granular degeneration and steatosis were observed; the islet atrophy, the structure was diminished, the number of the cells was reduced, the disordered arrangement, cytoplasm swelling and vacuolar degeneration were found in both groups (P <0.05). The morphology and structure of liver tissue and islet cells in phycocyanin group were clear, the number of normal cells was increased, the arrangement was more regular, the number of swelling and degeneration cells was less and the number of apoptotic cells was higher than that of normal cells The model group decreased significantly (P <0.05). Conclusion Phycocyanin can inhibit hepatic and pancreatic islet cell apoptosis in diabetic rats and play a protective role.
