目的探讨浒苔多糖对大鼠的血脂影响及抗脂质过氧化作用。方法利用高脂饲料建立高脂血症模型,给予不同剂量的浒苔多糖进行干预,测定血脂水平、计算动脉粥样硬化指数(AI),同时测定超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性及丙二醛(MDA)含量,并切片观察大鼠肝脏细胞。结果低、中、高剂量浒苔多糖均可显著降低高脂血症实验大鼠的血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)和丙二醛含量(P<0.01),其最大降幅分别为41.7%,79.3%,59.6%和55.0%;中、高剂量浒苔多糖可显著降低AI(P<0.01),其最大降幅为79.7%;低、中、高剂量浒苔多糖均可显著提高高密度脂蛋白胆固醇(HDL-C)含量和SOD、GSH-Px活性(P<0.01),其最大增幅分别为66.6%,63.8%和48.9%;同时,浒苔多糖还可降低高脂血症大鼠肝细胞的脂肪变性程度。结论浒苔多糖对实验性高血脂症模型大鼠具有降血脂、抗脂质过氧化及预防脂肪肝的作用。
Objective To investigate the effect of polysaccharides from Enteromorpha prolifera on lipid metabolism and anti-lipid peroxidation in rats. Methods Hyperlipidemia model was established using high-fat diet. Different doses of Enteromorpha polysaccharides were intervened to measure lipid levels, calculate atherosclerosis index (AI), and measure superoxide dismutase (SOD) and glutathione simultaneously. Peptide peroxidase (GSH-Px) activity and malondialdehyde (MDA) content were observed and liver cells were sectioned. Results Low, medium and high doses of polysaccharides from Enteromorpha prolifera could significantly reduce serum total cholesterol (TC), triglyceride (TG), low density lipoprotein cholesterol (LDL-C) and malondialdehyde in experimental hyperlipidemia rats. The content (P<0.01) had the largest reductions of 41.7%, 79.3%, 59.6%, and 55.0%, respectively. The middle and high doses of polysaccharides from Enteromorpha had a significant decrease in AI (P<0.01), with a maximum decrease of 79.7%. The medium and high doses of polysaccharides from Enteromorpha prolifera could significantly increase the levels of high-density lipoprotein cholesterol (HDL-C) and SOD and GSH-Px (P<0.01), with the maximum increases of 66.6%, 63.8%, and 48.9%, respectively. The polysaccharide from Enteromorpha may also reduce the steatosis of hepatocytes in hyperlipidemic rats. Conclusion The polysaccharides from Enteromorpha prolifera can inhibit blood lipids, resist lipid peroxidation and prevent fatty liver in experimental hyperlipidemia rats.
