通过受精卵原核显微注射的途径,产生带有可诱导型丙肝病毒非结构蛋白3基因(nonstructural3gene)的重组载体的转基因小鼠。采用PCR和基因组Southernblot杂交方法,在21只新生鼠中筛选到3只建立者小鼠。其中2只与非转基因小鼠进行交配,分别得到8只和6只F1代小鼠。进而对其基因组DNA进行检测,发现其中4只为转基因阳性个体。这些小鼠的获得,为ns3转基因品系的建立和NS3蛋白的生物学作用研究准备了条件。
Transgenic mice with a recombinant vector of inducible hepatitis C virus nonstructural 3 gene were generated by pronuclear microinjection of fertilized eggs. Three founder mice were screened from 21 neonatal rats by PCR and Southern blot hybridization. Two of them were mated with non-transgenic mice, and 8 and 6 F1 mice were obtained respectively. And then its genomic DNA was detected and found that 4 of them were transgenics positive individuals. The availability of these mice prepared the conditions for establishing a ns3 transgenic line and studying the biological role of the NS3 protein.
