目的 :揭示人绒毛膜促性腺激素 (hCG)是否通过影响细胞因子的生成而改变滋养层细胞或滋养层细胞来源的绒癌细胞的侵袭性。方法 :以JEG 3绒癌细胞系为研究对象 ,采用半定量逆转录多聚酶链反应 (RT PCR)方法 ,观察了hCG对JEG 3细胞表达胰岛素样生长因子 II(IGF II)的影响。结果 :分别用 0、5 0、5 0 0、5 0 0 0、2 5 0 0 0mIU/mLhCG处理 4 8h后 ,JEG 3细胞中IGF IImRNA的含量逐渐降低 ,并表现出一定的浓度效应———随hCG作用浓度增高而显著 ;另外 ,观察受 2 5 0 0 0mIU/mLhCG处理0、6、12、2 4、30、36、4 8h后JEG 3细胞中IGF II表达的变化情况 ,发现IGF II的表达并非一直受到抑制 ,而是在经过短暂的诱导后再降低。结论 :hCG可能通过调节IGF II在滋养层细胞或滋养层细胞来源的绒癌细胞中的表达 ,进而影响细胞的侵入或浸润转移
OBJECTIVE: To reveal whether human chorionic gonadotropin (hCG) alters the invasiveness of trophoblastic or trophoblast-derived choriocarcinoma cells by affecting cytokine production. Methods: The effects of hCG on the expression of insulin-like growth factor II (IGF II) in JEG 3 cells were observed by semi-quantitative reverse transcriptase-polymerase chain reaction (RT PCR) in JEG 3 choriocarcinoma cell line. Results: After treated with 0, 50, 50, 50, 0, 25 000 micromol / mL hCG for 48 h, the levels of IGF II mRNA in JEG 3 cells gradually decreased and showed a certain concentration effect - - significantly increased with increasing concentration of hCG. In addition, IGF II expression in JEG 3 cells treated with 25,000 micromol / L HCG at 0, 6, 12, 24, 30, 36 and 48 h was observed. IGF The expression of II was not always suppressed, but decreased after a brief induction. Conclusion: hCG may affect the invasion or metastasis of cells by regulating the expression of IGF II in choriocarcinoma cells derived from trophoblast or trophoblast cells
