目的建立测定地黄寡糖中地黄苷A的高效液相色谱法。方法采用Agilent TC C18色谱柱(4.6 mm×250 mm,5μm),柱温35℃,流动相:甲醇-水(10∶90),流速1.0 mL·min-1;ELSD参数:漂移管温度110℃,载气流量3.2 L·min-1。结果地黄苷A在0.2~1.2 mg·mL-1(r=0.999 3)内线性关系良好,平均回收率为98.8%(RSD=1.6%)。结论该方法准确、简便、专属性好,可用于地黄寡糖中地黄苷A成分的含量测定,为地黄寡糖的质量控制提供了依据。
Objective To establish a HPLC method for the determination of rehmannin A in Rehmannia glutinosa oligosaccharides. Methods The mobile phase was methanol-water (10: 90) at a flow rate of 1.0 mL · min-1 with an Agilent TC C18 column (4.6 mm × 250 mm, 5 μm) at 35 ℃. ELSD parameters: drift tube temperature of 110 ℃ , Carrier gas flow 3.2 L · min-1. RESULTS Rehumoside A had a good linearity within 0.2-1.2 mg · mL-1 (r = 0.999 3) with an average recovery of 98.8% (RSD = 1.6%). Conclusion The method is accurate, simple and specific. It can be used to determine the content of rehmannia glycoside A in Rehmannia glutinosa oligosaccharides and provide the basis for the quality control of Rehmannia glutinosa oligosaccharide.
