Proteins exert their biological functions and usually interact with other proteins. These interacting regions are called “hot spots”. Some abnormal interactions may lead to some diseases. Some specific The structure of small molecule drugs can inhibit these interactions, and thus achieve the purpose of treatment of disease. This paper reviews the constitution of the hot spots in protein-protein interactions (PPIs), the relationship between the variation of hot spots and diseases, the prediction of hot spots, As well as the interaction of several “hot spots” with small drug molecules to provide a reference for the development of small molecule drugs that regulate PPIs.
