对异体非骨化软骨细胞移植过程中免疫抑制的有效性进行了评价,比较了异体软骨细胞移植和整块软骨移植所引发的免疫反应。将成年雌鼠作宿主,5天龄幼雌鼠作供体,分别用与宿主相同及不同品系动物作为异体同系和异体异系移植。移植物取自肋软骨,经胶原酶消化后提取活的软骨细胞液(2.5×10~6个细胞/ml),存活率90%。整块肋软骨植入宿主背肌中,肋软骨细胞悬液0.1ml注入胫后肌肌腹内。免疫抑制剂联合用ATS和PCH,及单独用Cy-A,自移植当天起投药。所有动物在移植6周后宰杀,取移植组织及其周围组织作光镜观察。宿
The effectiveness of immunosuppression during allogeneic ossification chondrocyte transplantation was evaluated, and the immune responses induced by allogeneic chondrocyte transplantation and monolithic cartilage transplantation were compared. Adult female rats as the host, 5-day-old female mice as donors, respectively, with the same host and different strains of animals as allogeneic and allogeneic transplantation. The graft was taken from the costal cartilage, and after the collagenase digestion, the living chondrocyte fluid (2.5 × 10 ~ 6 cells / ml) was obtained and the survival rate was 90%. The entire costal cartilage implanted in the host back muscle, costal cartilage cells 0.1ml into the posterior tibialis muscle intramuscular injection. Immunosuppressive agents were administered in combination with ATS and PCH, and Cy-A alone, from the day of transplantation.
