应用随机扩增多态性DNA(RAPD)技术,对汝城早芽、蓝标1号、云桂大叶、日铸茶、举岩茶等5份茶树优质资源及其扦插繁殖后代的遗传稳定性进行了分析。结果表明,基因组DNA的平均得率为772μg/g鲜重,A260/A280在169~183之间,20个随机引物在5份资源中共扩增出412个位点,平均每个引物206个,每份资源824个位点,扩增产物片段大小约在04~25kb之间,多态性程度达849%。20个随机引物在5份优质资源及其扦插繁殖后代的RAPD扩增结果显示,无论是谱带的类型还是谱带的强弱,资源内是一致的,从而在DNA分子水平上证明了扦插繁殖的遗传稳定性。
