目的:研究羟基红花黄色素A(HSYA)对人肝癌细胞株(HepG2)培养上清液诱导的人脐静脉内皮细胞(HUVEC)增殖的抑制作用。方法:原代培养HUVEC,传至第3-6代进行实验。以MTT比色法检测不同浓度HSYA在不同作用时间,对HepG2培养上清液刺激后异常增殖的内皮细胞生长的抑制作用。结果:HSYA在0.0073g/L-0.00045g/L浓度范围内作用24h后,对于肿瘤上清液刺激后异常增殖的HUVEC有显著抑制作用(F=13.015,P<0.01);同样浓度范围作用24h后,对正常HUVEC生长无明显促进或抑制作用。结论:一定浓度的HSYA在一定时间范围内可以显著抑制肿瘤上清液刺激后异常增殖的人脐静脉内皮细胞。提示HSYA有可能成为一种新的肿瘤血管生成阻断剂。
AIM: To investigate the inhibitory effect of hydroxysafflor yellow A (HSYA) on the proliferation of human umbilical vein endothelial cells (HUVECs) induced by cultured human hepatoma cell line HepG2. METHODS: Primary cultured HUVECs were passaged to passages 3-6 for experiments. MTT assay was used to detect the inhibitory effect of HSYA at different concentrations on the proliferation of endothelial cells stimulated by HepG2 supernatant. Results: HSYA inhibited the proliferation of HUVECs after treated with tumor supernatant for 24 h (F = 13.015, P <0.01) after treated with 0.003 g / L-0.00045 g / L HSYA for 24 h After the normal HUVEC growth did not significantly promote or inhibit. Conclusion: A certain concentration of HSYA can significantly inhibit the proliferation of human umbilical vein endothelial cells stimulated by tumor supernatant within a certain period of time. Tip HSYA may become a new tumor angiogenesis blockers.
