研究橡胶种子在萌发进程中可溶性蛋白质含量及其电泳图谱,以及磷酸酶、过氧化物酶活性及其同工酶图谱的变化。结果表明:(1)子叶中可溶性蛋白质含量随种子萌发的进展而逐渐升高,其他部位的则下降。不同部位及不同萌发期的可溶性蛋白质电泳图谱的主要谱带基本不变,而弱谱带随部位及萌发期有较大的变化。(2)在贮藏期胚轴有较高的过氧化物酶活性;胚乳在各不同萌发期基本上不具有过氧化物酶活性,其他部位的过氧化物酶活性随萌发的进展而上升;其同工酶图谱也发生相应的变化。(3)随种子萌发的进展,各部位的酸性磷酸酶活性显著升高,其中子叶的变化尤为显著;碱性磷酸酶活性在贮藏期较弱,也随种子萌发而上升,但到胚很突出期后略有下降。磷酸酶同工酶图谱随种子萌发进展也发生消涨性变化。
The content of soluble protein and its electropherogram in rubber seeds during germination and the changes of phosphatase and peroxidase activities and their isozymes were studied. The results showed that: (1) The content of soluble protein in cotyledons increased gradually with the germination of seeds, but decreased in other parts. The main bands of soluble protein electrophoresis at different sites and different germination stages were basically the same, while the weak bands showed larger changes with site and germination. (2) The pericarp of pericarp had a higher peroxidase activity during storage; the endosperm basically had no peroxidase activity at different germination stages; the peroxidase activity of other parts increased with the progress of germination; Isozyme mapping also changes accordingly. (3) With the progress of seed germination, the acid phosphatase activity of all parts increased significantly, especially the cotyledon changes were particularly significant; alkaline phosphatase activity in the storage period is weak, also increased with the seed germination, but the embryo is very prominent After a slight decline. Phosphatase isoenzyme profile with the germination of the germplasm also occurred heave changes.
