目的:通过分析HL-60细胞内FLT3受体与STAT3的表达,探讨细胞信号转导异常与白血病的关系。方法:培养人早幼粒细胞白血病细胞株HL-60,利用特异性抗体,用免疫沉淀法、聚丙烯酰胺凝胶电泳及Western blot方法检测细胞内FLT3受体与STAT3的表达。结果:在HL-60细胞内检测到FLT3受体与STAT3的表达。结论:人早幼粒细胞白血病的发生、发展可能与酪氨酸激酶介导的细胞信号转导异常有关。
OBJECTIVE: To investigate the relationship between FLT3 receptor and STAT3 expression in HL-60 cells and the relationship between leukemia and cell signal transduction. Methods: The human promyelocytic leukemia cell line HL-60 was cultured and the expression of FLT3 receptor and STAT3 were detected by immunoprecipitation, polyacrylamide gel electrophoresis and Western blot using specific antibodies. Results: FLT3 receptor and STAT3 expression were detected in HL-60 cells. Conclusion: The occurrence and development of human promyelocytic leukemia may be related to the abnormal signal transduction mediated by tyrosine kinase.
