目的:观察膈下逐瘀汤对肝纤维化大鼠肝组织基质金属蛋白酶-9(MMP-9)及基质金属蛋白酶组织抑制因子-2(TIMP-2)表达的影响。方法:大鼠随机分为空白对照组、膈下逐瘀汤组、猪血清组、猪血清+膈下逐瘀汤组。猪血清腹腔注射诱导大鼠免疫性肝纤维化模型,同时膈下逐瘀汤灌胃给药干预。消化法检测肝组织羟脯氨酸含量,ELISA法检测肝组织MMP-9和TIMP-2蛋白含量,实时定量RT-PCR检测肝组织MMP-9和TIMP-2 mRNA表达。结果:与模型对照组比较,膈下逐瘀汤可显著降低肝组织羟脯氨酸、MMP-9及TIMP-2蛋白含量,下调肝组织MMP-9、TIMP-2 mRNA水平。结论:膈下逐瘀汤可有效地逆转实验性大鼠肝纤维化,其机制可能与调节MMP-9和TIMP-2的表达有关。
Objective: To observe the effect of Gexia Zhuyu Decoction on the expression of matrix metalloproteinase-9 (MMP-9) and tissue inhibitor of metalloproteinase-2 (TIMP-2) in hepatic tissue of hepatic fibrosis rats. Methods: The rats were randomly divided into blank control group, Gexia Zhuyu Decoction group, pig serum group and pig serum + Gexia Zhuyu Decoction group. The model of immunological liver fibrosis was induced by intraperitoneal injection of porcine serum, and the intervention of Gexia Zhuyu Decoction intragastrically was given. The content of hydroxyproline in liver tissue was detected by digestion method. The content of MMP-9 and TIMP-2 in liver tissue was detected by ELISA. The mRNA expression of MMP-9 and TIMP-2 in liver tissue was detected by real-time quantitative RT-PCR. Results: Compared with the model control group, Gexiazhuyutang significantly reduced the content of hydroxyproline, MMP-9 and TIMP-2 in liver tissue and down-regulated the mRNA levels of MMP-9 and TIMP-2 in liver tissue. Conclusion: Gexia Zhuyu Decoction can effectively reverse the experimental rat liver fibrosis, the mechanism may be related to the regulation of the expression of MMP-9 and TIMP-2.
