目的:研究桃红四物汤药物成分对血管平滑肌细胞(VSMC)FAK/P38MAPK信号表达的影响,并探讨其在抑制VSMC的迁移及增殖中的相关机理。方法:制备桃红四物汤含药血清,体外分离培养大鼠颈总动脉VSMC,将VSMC分为正常对照组(A组,正常大鼠血清培养)与干预组(B组,桃红四物汤含药血清干预)分别进行处理,采用免疫细胞化学方法检测各组FAK的表达情况,采用Western blot法检测各组不同时间及不同浓度含药血清干预下P38MAPK表达的变化。结果:大鼠VSMC在含药血清干预后,FAK及P38MAPK的表达均降低(P<0.01),P38MAPK的表达随含药血清干预时间的延长及作用浓度的增加而降低(P<0.01)。结论:桃红四物汤药物成分对FAK/P38MAPK信号的表达产生负性影响,此影响可能参与了对VSMC的迁移及增殖的抑制作用。
OBJECTIVE: To study the influence of the composition of Taohong Siwu Decoction on the expression of FAK / P38MAPK signal in vascular smooth muscle cells (VSMCs) and to explore its mechanism in inhibiting VSMC migration and proliferation. Methods: The serum of Taohong Siwu decoction was prepared. VSMC of rat common carotid artery was isolated and cultured in vitro. VSMC was divided into normal control group (A group, normal rat serum culture) and intervention group (B group, Taohongsiwu decoction The serum levels of FAK were detected by immunocytochemistry. The expression of P38MAPK in each group was detected by Western blot at different times and concentrations. Results: The expression of FAK and P38MAPK in rat VSMCs were decreased after intervention with serum (P <0.01). The expression of P38MAPK decreased with the prolongation of serum containing serum and the concentration of P38MAPK (P <0.01). Conclusion: The drug composition of Taohong Siwu decoction negatively affects the expression of FAK / P38MAPK signal, and this effect may be involved in the inhibition of VSMC migration and proliferation.
