目的 :HSV 1可引起多种疾病 ,ACV(阿昔洛韦 )治疗HSV 1感染效果虽好 ,但易出现耐药毒株。新药Cidofovir对ACV耐药毒株效果好 ,毒性也大。HSV疫苗研究主要针对生殖道疱疹 ,效果不好。为了解决这些问题 ,研制新的抗HSV中药方剂的最佳配比 ,我们进行了此项研究。方法 :以ACV为阳性对照 ,采用空斑减数试验 ,观察了虎杖金银花单用及联用抗HSV 1F株及HS 1株的药效 ,并且用中效作用原理对联用药效进行了分析。结果 :在HEp 2细胞系统中 ,当虎杖 /金银花是 1 (ED50 )虎杖 /2 (ED50 )金银花时 ,对F株及HS 1株有直接杀灭及感染的阻断作用 ,以及对F株及HS 1株增殖有抑制作用 ,当空斑减数率从 2 0 %~ 90 %时 ,CI<1 ,协同作用极为明显 ,这是首选联用比。结论 :虎杖与金银花以 1 (ED50 )虎杖 /2 (ED50 )金银花联用 ,对F株的治疗指数 (TI)为ACV的 4 60 7,2 87和4 2 5倍 ;对HS 1株TI为ACV的 548,3 93和 50 9倍。我们认为对于抗HSV 1而言 ,这是有利的药物联用组合。
Objective: HSV 1 can cause a variety of diseases, ACV (acyclovir) treatment of HSV 1 infection is good, but susceptible to drug-resistant strains. The new drug Cidofovir is effective against ACV-resistant strains and has high toxicity. The HSV vaccine study is mainly directed at the genital herpes and the effect is not good. In order to solve these problems and develop new anti-HSV prescriptions, we conducted this study. METHODS: Using ACV as positive control and plaque reduction experiment, the efficacy of single use of Polygonum cuspidatum honeysuckle alone and combined with anti-HSV 1F strain and HS 1 strain was observed, and the efficacy of the combined use was analyzed with the principle of intermediate effect. RESULTS: In the HEp 2 cell system, when Polygonum cuspidatum/Hydenthus was 1 (ED50) tussah / 2 (ED50) honeysuckle, the direct killing and infection of F strain and HS 1 strain was blocked, and the strain F and The proliferation of HS 1 strain was inhibited. When the plaque reduction rate was from 20% to 90%, the CI was less than 1 and the synergy was extremely obvious. This is the preferred combination ratio. Conclusion: Polygonum cuspidatum and honeysuckle were used in combination with 1 (ED50) knotweed/2 (ED50) honeysuckle, and the therapeutic index (TI) of the F strain was 4 60 7, 2 87 and 4 2 5 times that of ACV; TI of HS 1 strain was ACV 548, 3 93 and 50 9 times. We think that for anti-HSV 1, this is an advantageous drug combination combination.
