从韭菜花杆中提取分离出一种植物凝集素,对小鼠血红细胞有很强的凝集活性,当浓度为7.8μg·mL-1时,就能发生凝集反应。每100g的新鲜韭菜花杆经超声、离心、硫铵沉淀、透析、冻干后最终得0.099g粗品。考察韭菜凝集素提取过程中各工艺条件对提取分离的影响,采用蛋白质定量法评价各工艺参数的影响程度,建立最佳提取工艺,结果如下:超声时间为4min,离心转速为10000rpm,硫酸铵的饱和度为80%。
The lectin from leek flower stem isolated from a plant agglutinin has strong agglutination activity on mouse red blood cells, when the concentration of 7.8μg · mL-1, the agglutination can occur. Per 100g of fresh chives flower stem by ultrasound, centrifugation, ammonium sulfate precipitation, dialysis, freeze-dried finally get 0.099g crude. The effects of various technological conditions on the extraction and isolation of leek lectin during the extraction process were investigated. The effects of various technological parameters were evaluated by protein quantification. The optimal extraction process was established. The results were as follows: the ultrasonic time was 4 min, the centrifugal speed was 10000 rpm, Saturation is 80%.
