目的观察生存素(survivin)反义寡核苷酸(anti-survivin oligonucleotides,ASODN)对人胃癌细胞株BGC-823增殖、凋亡、侵袭、迁移能力的影响。方法脂质体介导ASODN或对照SODN转染人胃癌细胞株BGC-823后,RT-PCR方法检测survivin的mRNA表达水平,四氮唑盐(MTT)法检测ASODN或SODN对肿瘤细胞的增殖抑制效果,Transwell小室法检测细胞侵袭和迁移能力的改变,流式细胞术检测细胞周期及凋亡指数的改变。结果 ASODN转染可明显降低survivin的表达(P<0.05),并且ASODN转染具有以浓度及时间依赖的方式抑制BGC-823细胞增殖的作用(P<0.05)。用600 nmol/L浓度的ASODN转染BGC-823后,BGC-823细胞迁移和侵袭能力下降(P<0.05),细胞凋亡指数(apoptoticindex,AI)由(1.62±0.36)增加到(11.31±0.38)(P<0.01),而增殖指数(proliferation index,PI)则由(43.80±0.81)下降至(27.77±2.97)(P<0.05)。停滞于G2/M期的细胞明显增加(P<0.01)。结论 下调survivin表达的ASODN可通过抑制胃癌细胞增殖、诱导凋亡、降低胃癌细胞侵袭和迁移能力而发挥抗肿瘤作用。
Objective To investigate the effects of survivin ASODN on the proliferation, apoptosis, invasion and migration of human gastric cancer cell line BGC-823. Methods Liposome-mediated ASODN or control SODN was transfected into human gastric cancer cell line BGC-823. The mRNA expression of survivin was detected by RT-PCR. The proliferation of tumor cells was detected by MTT assay The changes of cell invasion and migration were detected by Transwell chamber assay. The cell cycle and apoptotic index were detected by flow cytometry. Results ASODN transfection could significantly decrease the expression of survivin (P <0.05), and ASODN transfection could inhibit the proliferation of BGC-823 cells in a concentration-and time-dependent manner (P <0.05). After transfection of BGC-823 with 600 nmol / L ASODN, the ability of migration and invasion of BGC-823 cells decreased (P <0.05) and the apoptotic index increased from (1.62 ± 0.36) to (11.31 ± 0.38) (P <0.01), while the proliferation index (PI) decreased from (43.80 ± 0.81) to (27.77 ± 2.97) (P <0.05). Cells arrested in G2 / M phase were significantly increased (P <0.01). Conclusion ASODN, which down-regulates the expression of survivin, can exert anti-tumor effects by inhibiting the proliferation and inducing apoptosis of gastric cancer cells and reducing the invasion and migration of gastric cancer cells.
