目的:研究紫草复方的水提取工艺条件和高效液相色谱法同时测定咖啡酸四聚体、丹酚酸B含量的方法。方法:采用正交试验设计法,分别考察加水量、煎煮时间、煎煮次数对咖啡酸四聚体、丹酚酸B含量和浸膏得率的影响,分别赋予咖啡酸四聚体、丹酚酸B含量和浸膏得率不同的权重系数(依次为0.4、0.4、0.2),进行多指标综合评分。结果:测定的流动相为甲醇∶0.5%磷酸水=40∶60;流速1ml/min,检测波长252nm。优化工艺条件为加16倍量的水,煎煮3次,每次1h。结论:高效液相色谱法同时测定咖啡酸四聚体、丹酚酸B含量的方法准确,线性好,该工艺稳定、简便。
Objective: To study the method of simultaneous determination of caffeic acid tetramer and salvianolic acid B by the water extraction process conditions and high performance liquid chromatography (HPLC) of comfrey compound. Methods: The effects of water addition, decocting time and boiling times on the content of caffeic acid tetramer, salvianolic acid B and the yield of extract were investigated by orthogonal design. The effects of caffeic acid tetramer, Phenolic acid B content and extract rate of different weight coefficients (followed by 0.4,0.4,0.2), multi-index composite score. Results: The mobile phase was methanol: 0.5% phosphoric acid water = 40:60; the flow rate was 1 ml / min and the detection wavelength was 252 nm. Optimize the process conditions plus 16 times the amount of water, boiling 3 times, each 1h. Conclusion: Simultaneous determination of caffeic acid tetramer and salvianolic acid B by HPLC method is accurate and linear. The method is stable and simple.
