目的探讨大鼠肝缺血-再灌流损伤时TNF-α、IL-1βmRNA在心肌组织中的表达及与心肌细胞凋亡的关系。方法选取健康雄性Wistar大鼠48只,随机分为6组,每组8只。应用免疫组织化学方法和原位杂交方法分别测定心肌组织中TNF-α、IL-1βmRNA的表达;应用原位细胞凋亡法测定细胞凋亡情况。同时观察心肌HE染色。结果I/R组与假手术组TNF-α在心肌组织中的表达,差异有显著性(P<0.01),I/R1h组明显升高,与I/R组比较差异有显著性(P<0.01),I/R2h组、I/R4h组虽然略有波动,但增加差异无显著性(P>0.05)。IL-1βmRNA自I/R组起表达就明显增多,与I组比较差异有显著性(P<0.01);I/R1h组明显升高,与I/R组比较差异有显著性(P<0.05)。细胞凋亡指数I/R组与假手术组比较明显增加(P<0.01),随着再灌流时间的延长细胞凋亡指数逐渐增加。TNF-α、IL-1βmRNA和细胞凋亡指数间均存在正相关(r=0.94,r=0.91;P<0.05)。HE染色后,见心肌部分横纹不清,有炎性细胞浸润,心肌纤维肿胀,心肌水肿。结论肝缺血-再灌流过程中,心肌组织早期就出现TNF-α和IL-1βmRNA的表达,心肌细胞凋亡指数随着再灌流时间的延长逐渐增加,可能与TNF-α和IL-1βmRNA的表达有关;心肌结构出现了病理改变。
Objective To investigate the expression of TNF-α and IL-1βmRNA in rat hepatic ischemia-reperfusion injury and its relationship with myocardial cell apoptosis. Methods Forty-eight healthy male Wistar rats were randomly divided into 6 groups with 8 rats in each group. The expression of TNF-α and IL-1βmRNA in myocardium was detected by immunohistochemistry and in situ hybridization, respectively. The apoptosis of cells was detected by in situ cell apoptosis assay. At the same time observe the myocardial HE staining. Results The expression of TNF-α in I / R group and sham operation group was significantly different from that in I / R group (P <0.01), and significantly increased in I / R1h group (P < 0.01). Although the I / R2h group and I / R4h group fluctuated slightly, the difference was not significant (P> 0.05). The expression of IL-1βmRNA in I / R group was significantly increased compared with that in I group (P <0.01); the I / R1h group was significantly higher than that in I / R group (P <0.05) ). The apoptotic index I / R group was significantly increased compared with the sham operation group (P <0.01), and the apoptosis index gradually increased with the reperfusion time prolonging. There was a positive correlation between TNF-α, IL-1βmRNA and apoptosis index (r = 0.94, r = 0.91; P <0.05). HE stained, see the myocardial part of the horizontal pattern unclear, inflammatory cell infiltration, myocardial fiber swelling, myocardial edema. Conclusions The expression of TNF-α and IL-1βmRNA in the early stage of myocardial ischemia-reperfusion is increased. The apoptotic index of myocardial cells gradually increases with the reperfusion time, which may be related to the changes of TNF-αand IL-1βmRNA Expression related; myocardial structure appeared pathological changes.
