目的探讨从蛋白及细胞水平检测人神经元特异性烯醇化酶(NSE)在非神经内分泌性肿瘤细胞系中的表达及其意义。方法用NSE特异引物和逆转录聚合酶链反应(RTPCR)、蛋白印迹、免疫细胞化学(ICC)染色技术检测NSE在肿瘤细胞中的表达。结果从转录蛋白及细胞水平都能从黑色素瘤细胞株WM451和WM983A、前列腺癌高转移细胞株PC3M和低转移细胞株PC3、乳腺癌细胞株MCF7、结直肠癌细胞株HT29和SL180、胃癌细胞株MGC803中检测到NSE的表达,阳性率为100%。结论NSE除在神经内分泌细胞表达外,在肿瘤细胞亦有更为广泛的表达谱,其作为神经内分泌来源肿瘤标志物的作用值得深入研究。
Objective To investigate the expression and significance of human neuron-specific enolase (NSE) in non-neuroendocrine tumor cell lines at the protein and cell level. Methods NSE specific primers and reverse transcriptase polymerase chain reaction (RTPCR), Western blot and immunocytochemistry (ICC) staining were used to detect the expression of NSE in tumor cells. Results From the transcriptional level and cellular level, all of the melanoma cell lines WM451 and WM983A, the prostate cancer cell line PC3M and the low metastasis cell line PC3, the breast cancer cell line MCF7, the colorectal cancer cell line HT29 and SL180, the gastric cancer cell line NSE expression was detected in MGC803 with a positive rate of 100%. Conclusion In addition to the expression of NSE in neuroendocrine cells, there is also a more extensive expression pattern in tumor cells. As a neuroendocrine tumor marker, NSE deserves further study.
