目的研究姜黄素对大鼠慢性心力衰竭心脏血管紧张素Ⅰ型受体(angiotensin Ⅱ type 1 receptor,AT1R)的表达作用。方法结扎大鼠左冠状动脉前降支制备心肌梗死后心衰模型,32只Wistar大鼠随机分为假手术组,假手术姜黄素处理组,心衰对照组及心衰姜黄素处理组。超声心动图检测各组大鼠心功能,放射免疫法测定血浆肾素(plasma renin actvity,PRA)及血管紧张素Ⅱ(angiotensionⅡ,AngⅡ)水平,qRT-PCR检测各组大鼠心脏AT1受体的mRNA表达,Western印迹法测定各组大鼠心脏AT1受体的蛋白表达。结果与假手术组相比,心衰对照组左心室射血分数显著降低(P<0.05),而血浆PRA及AngⅡ水平升高(P<0.05),同时心脏AT1受体mRNA及蛋白的表达水平也显著上调(P<0.05)。给予姜黄素处理4周后,可明显改善心衰大鼠上述指标的异常(P<0.05)。而姜黄素对假手术组无相应作用。结论姜黄素通过降低心衰大鼠心脏AT1受体的表达,改善慢性心衰大鼠心功能。
Objective To investigate the effect of curcumin on the expression of angiotensin Ⅱ type 1 receptor (AT1R) in chronic heart failure rats. Methods The left anterior descending coronary artery was ligated to establish the model of heart failure after myocardial infarction. Thirty-two Wistar rats were randomly divided into sham-operation group, curcumin-treated group, heart failure control group and curcumin treatment group. Cardiac function was measured by echocardiography, plasma renin activity (PRA) and angiotensin Ⅱ (AngⅡ) levels were measured by radioimmunoassay. The expression of AT1 receptor in heart of each group was detected by qRT-PCR The mRNA expression of AT1 receptor was detected by Western blotting. Results Compared with the sham-operated group, the left ventricular ejection fraction decreased significantly (P <0.05) and the plasma PRA and AngⅡ levels increased (P <0.05), while the expression of AT1 receptor mRNA and protein Also significantly up-regulated (P <0.05). Curcumin treatment for 4 weeks, can significantly improve the abnormal heart failure rats above indicators (P <0.05). Curcumin had no corresponding effect on the sham-operated group. Conclusion Curcumin improves heart function in rats with chronic heart failure by decreasing the expression of AT1 receptor in heart of rats with heart failure.
