作者在ph阳性的慢性粒细胞白血病(CGL)患者血液中发现存在有一种嗜酸和嗜硷性颗粒的混合型粒细胞.为明确同时存在这二种颗粒,作者建立了一种能鉴别嗜酸、嗜硷性颗粒的复染法,即对嗜硷性颗粒特异的甲苯胺兰(Toluidine blue)染色和对嗜酸性颗粒特异的抗氰化物过氧化酶联合染色法.方法:将空气干燥的血片和髓片先浸入pH6.6冷福马尔丙铜液配制的10%甲苯胺兰染色液中20分分钟后,用自来水冲洗2分钟;再浸入含有pH7.4的0.067moL/L磷酸盐缓冲液50ml;3.3一二氨基联苯胺37.5mg;30g/L过氧化氢0.15mL;氰化钠19.6mg的混合液中,置室温下10分钟后用自来水冲洗,自然干躁后加Permount盖片镜检.嗜酸性颗粒染色呈绿黄色,嗜硷性颗粒染紫黑色.作者应用梅-姬氏复染法(Mag-Grunwald-Giemsa)首先在一例CGL未稍血片中发现存在嗜酸、嗜硷性颗
