分别应用免疫组织化学和PCRSSCP方法检测14 例卵巢恶性肿瘤中p16 蛋白的表达及p16 基因外显子1 和外显子2 基因突变,其中5 例还同时检测转移灶癌组织。结果示7 例卵巢恶性肿瘤组织为p16 蛋白表达阴性;无一例有p16 基因外显子1 突变;4 例有p16 基因外显子2 突变,其中2 例无p16 蛋白表达;5 例转移灶癌组织的基因突变和蛋白质表达与原发灶一致。提示部分卵巢恶性肿瘤的发生和发展与p16 基因的失活有关,基因突变是其中一种失活方式;卵巢恶性肿瘤中p16 基因突变发生在转移前,而非转移后
Immunohistochemistry and PCRSSCP method were used to detect the expression of p16 protein and mutations of p16 gene exon 1 and exon 2 in 14 cases of ovarian cancer. 5 of them were also detected in metastatic cancer tissues. The results showed that 7 cases of ovarian cancer tissue p16 protein expression was negative; no case of p16 gene exon 1 mutation; 4 cases of p16 gene exon 2 mutations, of which 2 cases without p16 protein expression; 5 cases of metastatic cancer tissue The gene mutation and protein expression consistent with the primary tumor. Suggesting that the occurrence and development of some ovarian malignancies are related to the inactivation of p16 gene. Mutation is one of the ways of inactivation.
