目的:建立鼻咽癌(nasopharyngeal carcinoma,NPC)蛋白质表达谱,为NPC蛋白质组学研究奠定基础。方法:应用激光捕获显微切割(laser capture microdissection,LCM)技术从NPC组织纯化NPC细胞,应用二维凝胶电泳(2-DE)分离LCM纯化NPC细胞的蛋白质,基质辅助激光解吸电离飞行时间质谱(MALDI-TOF-MS)鉴定蛋白质,利用生物信息学资源构建NPC的2-DE蛋白质组数据库。结果:建立了NPC蛋白质2-DE参考图谱,2-DE图谱共显示(1312±30)个蛋白质点,共鉴定了427个蛋白质点,包括241个非冗余蛋白质,并构建了NPC的2-DE蛋白质组数据库,这些数据可从本实验室网站(http://www.xyproteomics.org)上进行查询。结论:首次建立了LCM纯化NPC细胞2-DE蛋白质表达谱,为NPC蛋白质组学研究提供了重要的信息和资料。
OBJECTIVE: To establish the protein expression profile of nasopharyngeal carcinoma (NPC), which lays a foundation for the research of NPC proteomics. METHODS: NPC cells were purified from NPC tissues by laser capture microdissection (LCM). Proteins purified from NPC cells by 2-DE were separated by 2-DE, and matrix-assisted laser desorption / ionization time of flight mass spectrometry (MALDI-TOF-MS) was used to identify the proteins, and bioinformatics resources were used to construct NPC 2-DE proteome database. Results: The NPC protein 2-DE reference map was established. A total of 1312 ± 30 protein spots were found in the 2-DE map. A total of 427 protein spots were identified, including 241 non-redundant proteins, and NPC 2- DE proteome database, which is available on our lab website (http://www.xyproteomics.org). CONCLUSION: The 2-DE protein expression profile of LCM purified NPC cells was established for the first time, which provided important information and information for NPC proteomics research.
