目的了解基质细胞衍生因子-1α(SDF-1α)在皮质发育畸形(MCD)大鼠海马中的表达,探讨其在皮质发育畸形异常局部神经回路中的作用。方法通过腹部X射线照射孕16dSD大鼠构建子代MCD模型,对30、60d对照组与实验组子鼠(P30、P60)行HE、Timm染色以检测其组织学改变,免疫组化、RT-PCR及Western-Blot检测SDF-1α的表达情况。结果①Timm染色示模型鼠海马出现苔藓纤维出芽;②免疫组化示SDF-1α在模型组海马CA1区异位神经元有表达;③RT-PCR和Western-Blot结果显示模型组P30、P60较同期的对照组表达降低。结论 SDF-1α的下调可能参与皮质发育畸形海马中异常神经回路的形成。
Objective To investigate the expression of stromal cell-derived factor-1α (SDF-1α) in the hippocampus of cortical deformity (MCD) rats and to explore the role of SDF-1α in abnormal local neural circuits of cortical dysplasia. Methods MCD model of offspring was established by abdominal X-ray irradiation in pregnant SD rats. HE and Timm staining were used to detect the histological changes, immunohistochemistry and RT-PCR in 30-60 days control group and experimental group (P30, P60) PCR and Western-Blot detection of SDF-1α expression. Results ①There was moss fiber sprouting in the hippocampus of rats induced by Timm staining; ②Expression of SDF-1α in eutopic neurons of hippocampal CA1 area of the model group was observed by immunohistochemistry; ③The expression of P30 and P60 in the model group was significantly higher than that in the model group by RT-PCR and Western- The control group decreased expression. Conclusion The down-regulation of SDF-1α may be involved in the formation of abnormal neural circuits in cortical deformity hippocampus.
