目的研究木犀草素对UW液低温保存离体大鼠心脏的功能及对心肌细胞凋亡的影响。方法将40只成年SD大鼠随机分成2组(n=20):对照组(UW组)及实验组(添加30μmol/L木犀草素的UW液)。利用Langendorff离体心脏灌流法,观察离体心脏冷保存18h复灌60min后心率、左心室最大收缩压、左心室压力变化率、心肌细胞凋亡及凋亡相关基因Bax、Bcl-2的表达。结果与对照组比较,实验组复灌期心脏的收缩功能(LVPSP,+dp/dtmax)、舒张功能(-dp/dtmax)及心率高于对照组差异有统计学义意义(P<0.05);心肌细胞的凋亡及促进凋亡Bax基因的表达均明显低于对照组;而抑制凋亡Bcl-2基因的表达均明显高于对照组。结论木犀草素能显著改善UW液低温保存离体大鼠心脏的功能,对心肌细胞凋亡有明显抑制作用。
Objective To investigate the effect of luteolin on the function of isolated rat heart and its effect on cardiomyocyte apoptosis in UW solution. Methods Forty adult SD rats were randomly divided into two groups (n = 20): control group (UW group) and experimental group (UW solution supplemented with 30μmol / L luteolin). The heart rate, the maximum systolic pressure of left ventricle, the rate of change of left ventricular pressure, the apoptosis of cardiomyocytes and the expression of Bax and Bcl-2 were observed by Langendorff perfusion in vitro. Results Compared with the control group, the systolic function (LVPSP, + dp / dtmax), diastolic function (-dp / dtmax) and heart rate in the experimental group during reperfusion period were significantly higher than those in the control group (P <0.05). The apoptosis of cardiomyocytes and the expression of apoptosis-promoting Bax gene were significantly lower than those of the control group, while the expression of Bcl-2 gene was significantly higher than that of the control group. Conclusions Luteolin can significantly improve the function of UW fluid cryopreserved in isolated rat heart and significantly inhibit cardiomyocyte apoptosis.
