通过中试试验确定了姬松茸液体发酵生产菌丝体的最佳条件和菌丝体多糖生产工艺。发酵工艺中碳源为玉米粉和蔗糖,氮源为豆饼粉和酵母粉,接种量5%,发酵温度25℃,通气量1∶0.5(v/v),搅拌速度150 r/min,发酵培养72 h,菌丝体平均得率16.42%,菌丝体中多糖含量为25.22%。液体发酵姬松茸菌丝体多糖的生产工艺为:菌丝体加10倍量水,100℃热回流2次(提取时间分别为2 h和1 h),合并2次提取滤液,用截留值为10 kD的中空纤维超滤柱将提取液超滤浓缩至浓缩比为1∶1,将浓缩滤液加2%麦芽糊精,喷雾干燥后得粗多糖,粗多糖得率为12.64%,粗多糖含量为33.47%。
The optimal conditions for mycelium production by Agaricus blazei Murrill and the production technology of mycelium polysaccharide were determined through pilot plant experiments. The carbon sources in the fermentation process were corn flour and sucrose, the nitrogen source was soybean meal and yeast powder, the inoculation amount was 5%, the fermentation temperature was 25 ℃, aeration rate was 1: 0.5 (v / v) and stirring speed was 150 r / min. 72 h, average mycelial yield was 16.42%, polysaccharide content in mycelium was 25.22%. Liquid Agaricus Agaricus mycelia liquid polysaccharide production process: mycelium plus 10 times the amount of water, 100 ℃ heat reflux 2 times (extraction time were 2 h and 1 h), the combined extract was twice, with a cut-off value of 10 kD hollow fiber ultrafiltration column extract ultrafiltration concentrated to a concentration ratio of 1: 1, the filtrate was concentrated and 2% maltodextrin spray dried to give crude polysaccharide, crude polysaccharide yield was 12.64%, the crude polysaccharide content 33.47%.
