目的检测磷酸化小窝蛋白-1(PY14Cav-1)在食管癌细胞中的表达,探讨其表达的意义。方法体外培养人食管癌株TE1细胞,检测TE1细胞中PY14Cav-1的表达水平,通过细胞转染技术以Cav-1小干扰RNA转染TE1细胞,使细胞中PY14Cav-1表达水平降低,用Transwell细胞体外侵袭实验观察细胞侵袭情况。结果转染Cav-1siRNA后,食管癌细胞TE1中Cav-1和PY14Cav-1表达水平均较转染前明显下降(P均<0.05),而Transwell结果显示,转染Cav-1 siRNA后,细胞侵袭数目为(7.4±1.2)个,较转染前[(49.2±3.6)个]明显减少(P<0.01)。结论Cav-1在食管癌的发生发展过程中扮演着促癌基因的角色,其机制可能与调节PY14Cav-1的表达有关。
Objective To detect the expression of phosphorylated caveolin-1 (PY14Cav-1) in esophageal cancer cells and to explore its significance. Methods Human esophageal cancer cell line TE1 was cultured in vitro. The expression of PY14Cav-1 was detected in TE1 cells. The expression of PY14Cav-1 was reduced by Cav-1 siRNA in vitro. The expression of PY14Cav- In vitro cell invasion assay observed cell invasion. Results After Cav-1 siRNA transfection, the expression levels of Cav-1 and PY14Cav-1 in esophageal cancer TE1 cells were significantly decreased compared with that before transfection (P <0.05), while the results of Transwell assay showed that Cav-1 siRNA transfected cells The number of invasion was 7.4 ± 1.2, which was significantly lower than that before transfection (49.2 ± 3.6) (P <0.01). Conclusions Cav-1 plays a role of oncogene in the development and progression of esophageal cancer, and its mechanism may be related to the regulation of PY14Cav-1 expression.
