从核基质蛋白成分的改变探讨难治性白血病的机制;用高盐法抽提出细胞核基质蛋白,SDS-PAGE电泳法分析核基质蛋白的变化;结果显示对HAT化疗方案敏感的白血病患者的白血病细胞核基质蛋白成分明显消失或减少,而对HAT不敏感的白血病患者的白血病细胞核基质细胞蛋白成分变化不大;提示:白血病细胞中分子量为38、42、50、51、60、65、80kd的核基质蛋白成分可能与难治性白血病的发生机制有关
The mechanism of refractory leukemia was explored from the changes of nuclear matrix protein components; nuclear matrix proteins were extracted by high salt method, and the changes of nuclear matrix proteins were analyzed by SDS-PAGE electrophoresis; the results showed that leukemia cell nucleus was sensitive to HAT chemotherapy in leukemia patients. The protein content of matrix proteins disappeared or decreased obviously, while the protein content of leukemia cell nucleus stromal cells in leukemia patients with insensitivity to HAT did not change much. Note: The nuclear matrices with molecular weights of 38, 42, 50, 51, 60, 65, and 80 kd in leukemia cells. Protein composition may be related to the mechanism of refractory leukemia
