目的:观察辛伐他汀对实验性大鼠脑出血(intracerebral hemorrhage,ICH)损伤中凋亡相关因子肿瘤坏死因子-α(TNF-α)表达的影响,探讨其通过抗凋亡机制发挥的脑保护作用。方法:采用注入新鲜自体不凝血复制脑出血模型,干湿重法检测脑组织含水量;免疫组化法检测TNF-α和caspase-3的表达,Western Blot检测脑组织TNF-α和caspase-3的蛋白表达水平。结果:大鼠脑出血后血肿周围基底节区脑组织TNF-α和caspase-3凋亡相关蛋白表达量、脑组织含水量及凋亡细胞数量明显上调。与模型组相比,辛伐他汀干预组的凋亡细胞数、TNF-α和caspase-3的表达与脑组织损伤周围含水量均显著降低(P<0.05)。结论:辛伐他汀对大鼠脑出血损伤有一定的保护作用,可能通过抑制脑组织中TNF-α和caspase-3的表达,进而减少细胞凋亡。
OBJECTIVE: To observe the effect of simvastatin on the expression of tumor necrosis factor-α (TNF-α) in the experimental rat model of intracerebral hemorrhage (ICH) injury and to explore the protective effects of simvastatin on the brain via anti-apoptotic mechanism effect. Methods: The model of intracerebral hemorrhage was established by injecting fresh autologous blood without coagulation. The water content of brain tissue was measured by wet-dry method. The expressions of TNF-α and caspase-3 were detected by immunohistochemistry. The expressions of TNF-α and caspase-3 Protein expression level. Results: After intracerebral hemorrhage, the expression of TNF-α and caspase-3 protein, the water content of brain tissue and the number of apoptotic cells in the brain tissue around the hematoma were significantly increased. Compared with the model group, the apoptotic cells, the expression of TNF-α and caspase-3 in the simvastatin treatment group were significantly decreased (P <0.05). Conclusion: Simvastatin has a protective effect on intracerebral hemorrhage in rats, which may be related to the decrease of apoptosis by inhibiting the expression of TNF-α and caspase-3 in brain tissue.
