目的:观察宁痫颗粒联合卡马西平对耐药性癫痫大鼠即早基因c-fos表达的影响,并探讨其作用机制。方法:Wistar雄性大鼠42只,按体质量随机分为假手术组,模型组,卡马西平+宁痫颗粒高[(0.03+4.80)g.kg-1.d-1]、中[(0.03+2.40)g.kg-1.d-1]、低剂量组[(0.03+1.20)g.kg-1.d-1],宁痫颗粒组(2.40 g.kg-1.d-1)和卡马西平组(0.03.g.kg-1.d-1),每组6只。采用海人酸海马CA3局部注射制作大鼠癫痫模型,术后第2天予苯妥英钠药物筛选14 d后制作耐药性癫痫模型。各组灌胃2周后免疫组织化学法检测大鼠即早基因c-fos表达的影响。结果:免疫组化实验显示:各治疗组大鼠耐药性癫痫模型颞叶皮层、海马CA3区c-fos阳性细胞较假手术组表达增强,密集分布,染色较深,同时又低于模型组;宁痫颗粒联合卡马西平各组均抑制c-fos蛋白的表达,作用优于卡马西平组(P<0.05)。各组间比较无显著差异性,宁痫颗粒组与卡马西平组无统计学差异。结论:宁痫颗粒联合卡马西平能抑制即早基因c-fos表达,两者联合疗效明显增强,以联合宁痫颗粒高剂量组疗效最佳,但治疗组指标均完全恢复到假手术组水平。
Objective: To observe the effect of Ningxue granule combined with carbamazepine on the expression of c-fos, an early gene of drug-resistant epilepsy rats, and to explore its mechanism. Methods: Forty-two Wistar male rats were randomly divided into sham-operation group, model group, carbamazepine and nong-sipian granule [(0.03 + 4.80) g.kg-1.d-1] 0.03 + 2.40) g.kg-1.d-1], low-dose group [(0.03 + 1.20) g.kg-1.d- ) And carbamazepine group (0.03.g.kg-1.d-1), 6 in each group. The rat epilepsy model was made by local injection of kainate hippocampal CA3. The model of drug-resistant epilepsy was made on the second day after phenytoin sodium drug screening for 14 days. The expression of c-fos, an early gene of rat, was detected by immunohistochemistry in each group after 2 weeks. Results: Immunohistochemistry showed that the expression of c-fos positive cells in the temporal cortex and hippocampal CA3 region of the drug-resistant epilepsy model in each treatment group was stronger than that in the sham-operation group, densely distributed, deeply stained, and lower than the model group ; Ningxin Granule combined with carbamazepine all inhibited the expression of c-fos protein, which was better than carbamazepine group (P <0.05). There was no significant difference among the groups, and there was no significant difference between Ningxue Granule group and carbamazepine group. Conclusion: Ningxue granule combined with carbamazepine can inhibit the expression of immediate early gene c-fos, the combined effect was significantly enhanced in combination with ningxin Granules high-dose group the best effect, but the treatment group indicators were completely restored to sham operation group level .
