本文改进了扑尔敏血药浓度的气相测定法。选用安定为内标,在异辛烷一次提取全血后用N~(63)电子捕获器检测。结果表明,在0.5~12ng/ml范围内,扑尔敏及安定的色谱峰面积之比(Ac/Ad)与血液中两者浓度之比(Cc Cd)有良好的线性关系(r=0.996),回收率在90%以上(平均偏差4%)。应用此法测定了人口服扑尔敏微丸及速释微丸的血药浓度,并由此计算得出控释微丸的体内吸收百分数(Pi)与体外溶出百分数(Po)间的相关方程式为Po=0.9597Pi+3.3225(r=0.916)
This article improves the concentration of chlorpheniramine gas phase determination. Diazepam was used as an internal standard, and once the whole blood of isooctane was extracted, it was detected by N 63 electron capture device. The results showed that there was a good linear relationship (r = 0.996) between the ratio of peak area of chlorpheniramine and diazepam (Ac / Ad) in the range of 0.5-12 ng / ml and the concentration of Cc Cd in blood , The recovery rate of more than 90% (average deviation of 4%).
