为探讨纤维连接蛋白(FN)及其β1整合素受体在人胎儿和成熟肾中的表达及意义,采用免疫组化的方法观察纤维连接蛋白及其整合素受体(α5β1)的α5亚单位在胎儿和正常成熟肾组织中的分布。结果发现:在胚胎肾组织中,FN及α5在输尿管芽细胞周围有弱阳性表达,在肾小球发育早期,FN及α5在肾单位中表达较弱,其在间质网状分布,当血管插入肾单位后,FN表达明显增加,主要分布于系膜区和毛细血管壁。成熟肾组织中FN及α5在肾小球基底膜(GBM)强阳性表达且以血管极节段明显,间质及小血管壁亦为阳性。结果提示,FN和整合素的表达和相互作用在肾小球系膜区,肾间质及GBM的形成、发育过程中发挥重要作用
To investigate the expression of fibronectin (FN) and its β1 integrin receptor in human fetal and mature kidney and its significance, immunohistochemistry was used to observe the expression of α5 subunit of fibronectin and its integrin receptor (α5β1) In fetal and normal mature kidney tissue distribution. The results showed that in the embryonic kidney tissue, FN and α5 were weakly positive around the sprouting of the ureter. In the early stage of glomerular development, FN and α5 were weakly expressed in the nephron, which distributed in the interstitial network. After nephron insertion, FN expression increased significantly, mainly in the mesangial area and the capillary wall. Mature renal tissue FN and α5 in the glomerular basement membrane (GBM) strong positive expression and vascular segments were obvious, interstitial and small vessel wall is also positive. The results suggest that the expression and interaction of FN and integrin play an important role in the formation and development of glomerular mesangial area, renal interstitium and GBM
