目的:观察扶正祛邪中药复方含药血清对长春新碱诱导的急性早幼粒白血病耐药细胞株HL60/VCR细胞耐药基因Bcl-2表达水平的影响。方法:采用流式细胞术检测法,选择HL60/VCR细胞为靶细胞,观察不同浓度扶正祛邪含药血清对其耐药基因Bcl-2表达的影响。结果:扶正祛邪含药血清对HL60/VCR细胞内凋亡抑制基因Bcl-2的表达有明显抑制作用,其中含药血清高、中、低剂量组荧光强度依次为401.67±0.86、453.69±0.40、516.66±0.40。结论:扶正祛邪复方中药抗白血病多药耐药的作用可能与下调Bcl-2的表达有关。
Objective: To observe the effect of Fuzheng Quxie Chinese medicine serum on the expression of drug resistance gene Bcl-2 in vincristine-induced acute promyelocytic leukemia cell line HL60 / VCR. Methods: Flow cytometry was used to select HL60 / VCR cells as target cells. The effects of different concentrations of Fuzhengquyu serum on the expression of Bcl-2 gene were observed. Results: Fuzhengquxie containing serum had significant inhibitory effect on the expression of Bcl-2 in HL60 / VCR cells. The fluorescence intensities of high, medium and low dose groups were 401.67 ± 0.86, 453.69 ± 0.40 , 516.66 ± 0.40. Conclusion: The anti-leukemia multidrug resistance effect of Fuzhengquxue Compound Chinese herbal medicine may be related to the down-regulation of Bcl-2 expression.
