目的探讨热休克蛋白70在妊娠期高血压疾病孕妇血清、脐血及胎盘组织中的表达及其在发病中的作用。方法选取2013年1月-2014年4月间在西安医学院第二附属医院分娩的妊娠期高血压疾病孕妇105例,分为妊娠期高血压组、轻度子痫前期组,重度子痫前期组,每组各35例。选取同期在该院分娩的35例健康孕妇为对照组。采用ELISA法检测4组孕妇血清、脐血中的热休克蛋白70水平;采用免疫组织化SP法检测胎盘组织中热休克蛋白70蛋白表达水平;采用RT-PCR技术检测胎盘组织中热休克蛋白70 mRNA表达水平。结果轻度和重度子痫前期组孕妇血清和脐血中的热休克蛋白70水平均明显高于妊娠期高血压组和对照组,差异有统计学意义(均P<0.05);重度子痫前期组孕妇血清和脐血中热休克蛋白70水平明显高于轻度子痫前期组,差异有统计学意义(P<0.05);妊娠期高血压组与对照组孕妇之间差异无统计学意义(P>0.05)。妊娠期高血压组、轻度和重度子痫前期组胎盘组织中热休克蛋白70蛋白阳性率均明显高于对照组,差异有统计学意义(P<0.05);重度子痫前期组热休克蛋白70蛋白阳性率明显高于轻度子痫前期组和妊娠期高血压组,差异均有统计学意义(均P<0.05);轻度子痫前期组与妊娠期高血压组孕妇之间差异无统计学意义(P>0.05)。妊娠期高血压组、轻度和重度子痫前期组孕妇胎盘组织中热休克蛋白70 mRNA表达明显高于对照组,差异均有统计学意义(均P<0.05)。重度子痫前期组孕妇胎盘组织中热休克蛋白70 mRNA表达明显高于妊娠期高血压组和轻度子痫前期组,差异均有统计学意义(P<0.05)。结论妊娠期高血压疾病孕妇血清、脐血及胎盘组织中热休克蛋白70表达水平明显提升,且随病情严重程度加重而增加,提示热休克蛋白70在妊娠期高血压疾病病程中可能起到一定作用。
Objective To investigate the expression of heat shock protein 70 (HSP70) in serum, cord blood and placenta of pregnant women with hypertensive disorder of pregnancy and its role in the pathogenesis. Methods 105 pregnant women with gestational hypertension who gave birth in the Second Affiliated Hospital of Xi’an Medical College from January 2013 to April 2014 were divided into three groups: hypertensive pregnancy group, mild preeclampsia group, severe preeclampsia group, Group, 35 cases in each group. The levels of heat shock protein 70 in serum and umbilical cord blood of 4 pregnant women were detected by ELISA. Results The levels of HSP70 in serum and umbilical cord blood of pregnant women with mild and severe preeclampsia were significantly higher than those of gestational hypertension group and control group (all P <0.05). Severe preeclampsia The level of heat shock protein 70 in serum and umbilical cord blood of pregnant women was significantly higher than that of mild preeclampsia (P <0.05), and there was no significant difference between pregnant women with gestational hypertension and pregnant women P> 0.05). The positive rates of HSP70 in placental tissue of hypertensive pregnancy group and mild and severe preeclampsia group were significantly higher than those in control group (P <0.05). The levels of heat shock protein 70 protein in the mild preeclampsia group was significantly higher than that in the mild preeclampsia group and the gestational hypertension group (all P <0.05). There was no significant difference between the mild preeclampsia group and the gestational hypertension group Statistical significance (P> 0.05). The expression of heat shock protein 70 mRNA in placental tissue of gestational hypertension group and mild and severe preeclampsia group was significantly higher than that of control group (all P <0.05). The expression of heat shock protein 70 mRNA in placental tissue of pregnant women with severe preeclampsia was significantly higher than that of gestational hypertension and mild preeclampsia (P <0.05). Conclusions The expression of heat shock protein 70 in serum, umbilical cord blood and placenta of pregnant women with hypertensive disorder complicating pregnancy is significantly increased, and increases with the severity of the disease, suggesting that heat shock protein 70 may play a certain role in the course of hypertensive disorders of pregnancy effect.
