目的:探讨三氧化二砷(As_2O_3)对鸡胚移植胆管癌生长的抑制作用及其机制。方法:建立胆管癌QBC_(939)细胞株鸡胚移植模型,接种癌细胞后9d切取标本,比较As_2O_3作用组和对照组肿瘤生长、血管生成情况,原位末端标记法检测肿瘤组织细胞凋亡,免疫组化检测核增生抗原(PCNA)与血管内皮生长因子(VEGF)的表达。结果:As_2O_3作用组肿瘤生长和血管生成受到抑制,细胞凋亡增多、PCNA和VEGF表达减弱。结论:As_2O_3有抗胆管癌的作用,其机制与诱导细胞凋亡,抑制癌细胞增生、癌组织VEGF表达以及肿瘤血管生成有关。
Objective: To investigate the inhibitory effect of arsenic trioxide (As_2O_3) on the growth of cholangiocarcinoma and its mechanism. Methods: The chick embryo transplantation model of QBC 939 cell line was established. After inoculation of cancer cells, the specimens were taken 9 days after the inoculation. The growth and angiogenesis of As 2 O 3 -treated group and control group were compared. The apoptosis of tumor cells was detected by in situ end- Immunohistochemical detection of nuclear proliferative antigen (PCNA) and vascular endothelial growth factor (VEGF) expression. Results: The tumor growth and angiogenesis in As2O3-treated group were inhibited, the apoptosis increased, and the expression of PCNA and VEGF decreased. CONCLUSION: As2O3 has the anti-cholangiocarcinoma effect. Its mechanism is related to the induction of apoptosis, the inhibition of cancer cell proliferation, the expression of VEGF in cancer tissue and tumor angiogenesis.
