目的:探讨非洛地平对家兔血管成形术后内皮功能及内膜增殖的影响。方法:日本大耳白兔36只,高脂喂养4周后用球囊导管剥脱腹主动脉内皮,再高脂喂养4周后,对动脉粥样硬化部位行血管成形术,30只成功的动物随机分为对照组、高脂组和治疗组(非洛地平),继续喂养4周后采集局部血管行病理形态学分析,并观察实验前后各组血清脂质、一氧化氮及血浆内皮素-1的改变。结果:治疗组血管内膜面积与中膜面积之比及内膜面积均降低。各组高脂饮食后血清总胆固醇、甘油三酯和血浆内皮素-1均明显升高,而血清一氧化氮明显下降;12周治疗组血浆内皮素-1明显降低,而血清一氧化氮水平明显升高。结论:非洛地平具有抑制家兔血管成形术后内膜增殖和保护内皮功能的作用。
Objective: To investigate the effect of felodipine on endothelial function and intima proliferation after angioplasty in rabbits. Methods: Thirty-six Japanese white rabbits were sacrificed and balloon catheter was used to strip the abdominal aortic endothelium after 4 weeks of high fat diet. After 4 weeks of high fat diet, angioplasty was performed on the atherosclerotic plaques. Thirty successful animals The rats were randomly divided into control group, hyperlipidemia group and treatment group (felodipine). After 4 weeks of continuous feeding, local blood vessels were harvested for histopathological analysis. Serum lipids, nitric oxide and plasma endothelin- 1 changes. Results: The ratio of intima to media area and the intima area in the treatment group were decreased. Serum total cholesterol, triglyceride and plasma endothelin-1 were significantly increased in all groups after high-fat diet, while serum nitric oxide was significantly decreased; while plasma endothelin-1 in 12-week treatment group was significantly decreased, while serum nitric oxide Significantly increased. Conclusion: Felodipine can inhibit the intima proliferation and protect endothelial function after angioplasty in rabbits.
