目的 :检测丙酮酸羧化酶(pyruvate carboxylase,PC)在上皮性卵巢癌中的表达水平及其临床病理学意义,并进一步探讨沉默PC基因的表达对卵巢癌SKOV3细胞增殖及迁移的影响。方法 :分别采用蛋白质印迹法和免疫组织化学方法检测PC在上皮性卵巢癌及其相应的癌旁组织中的表达水平;统计分析PC表达与患者年龄、有无腹腔积液、临床TNM分期及病理分级等临床病理特征之间的关系。通过构建特异性针对PC基因的PC-shRNA沉默SKOV3细胞中PC的表达水平,并采用MTT法、二维克隆形成实验、软琼脂克隆形成实验及细胞迁移实验,观察沉默PC表达对SKOV3增殖及迁移能力的影响。结果 :PC在卵巢癌组织中的表达水平明显高于其对应的癌旁组织(t=20.4,P<0.01);PC蛋白高表达与患者发生腹腔积液和临床分期有关(P值均<0.05)。沉默PC的表达可以显著抑制SKOV3细胞的增殖和迁移能力(P值均<0.01)。结论 :PC在上皮性卵巢癌组织中高表达,PC与卵巢癌的发生和发展密切相关,可作为卵巢癌临床治疗的潜在靶点。
OBJECTIVE: To detect the expression of pyruvate carboxylase (PC) in epithelial ovarian cancer and its clinicopathological significance. To further investigate the effect of silencing PC gene expression on the proliferation and migration of ovarian cancer SKOV3 cells. Methods: The expression of PC in epithelial ovarian cancer and its corresponding paracancerous tissues was detected by Western blotting and immunohistochemistry respectively. The high expression of PC protein was associated with ascites and clinical stage (P <0.05) ). Silencing PC expression significantly inhibited the proliferation and migration of SKOV3 cells (P <0.01). Conclusions: PC is highly expressed in epithelial ovarian cancer. PC is closely related to the occurrence and development of ovarian cancer and may serve as a potential target for the clinical treatment of ovarian cancer.
