小牛软骨经盐酸胍抽提、丙酮分级沉淀及膜超滤等步骤获得了软骨抗肿瘤制剂(Carti-lage Antitumor Preparation, CATP)。利用~3H-TdR参入、测定肿瘤干细胞及细胞死亡率等方法研究了CATP对肿瘤细胞及血管内皮细胞的抑制效应,并对小鼠移植性肿瘤进行了抗肿瘤作用的研究。结果表明:(1)合适浓度的CATP可显著地抑制肿瘤细胞和血管内皮细胞的生长,而不抑制正常细胞;(2)CATP对荷瘤小鼠肿瘤具有明显的抑制作用,抑瘤率为50%~60%。给药组小鼠胸腺重量明显增加,提示CATP可能具有免疫调节作用。实验结果为CATP的临床应用提供了依据。
Cartilage cartilage was extracted with guanidine hydrochloride, fractionated by acetone and membrane ultrafiltrated to obtain Cartilage Antitumor Preparation (CATP). The inhibitory effect of CATP on tumor cells and vascular endothelial cells was studied by ~ 3H-TdR incorporation, the determination of tumor stem cells and cell death rate, and the anti-tumor effect of transplanted tumors in mice. The results showed that: (1) CATP significantly inhibited the growth of tumor cells and vascular endothelial cells without inhibiting normal cells; (2) CATP had a significant inhibitory effect on the tumor-bearing mice and the tumor inhibition rate was 50 % ~ 60%. The thymus weight of the mice in the treatment group was significantly increased, suggesting that CATP may have immunomodulatory effects. The experimental results provide the basis for the clinical application of CATP.
