目的:观察川崎病(Kawasaki disease,KD)合并冠脉损伤患儿血清对脐静脉内皮细胞(human umbilical vein endothelial cells,HUVECs)增殖及凋亡的影响,以进一步了解KD患儿血管内皮细胞损伤的机制。方法:HUVECs分为正常血清组(Control组)、发热血清组(F组)、KD无冠脉损伤组(Non-CALs组)和KD合并冠脉损伤组(CALs组)。MTT比色法检测细胞增殖,流式细胞术(flow cytometry,FCM)分析各组HUVECs的凋亡情况。结果:MTT比色结果显示,Non-CALs组和CALs组经血清干预后的细胞活性低于F组和Control组,且CALs组低于Non-CALs组,差异有统计学意义(P<0.01)。此外经KD患儿血清作用后,HUVECs的凋亡率增加,与对照组相比差异有统计学意义(P<0.01),且CALs组凋亡率高于Non-CALs组。结论:KD合并冠脉损伤患儿血清具有抑制HUVECs增殖并诱导HUVECs凋亡的作用。
OBJECTIVE: To observe the effects of serum of Kawasaki disease (KD) patients with coronary artery lesion on the proliferation and apoptosis of human umbilical vein endothelial cells (HUVECs) in order to further understand the effects of Kawasaki disease (KD) on vascular endothelial cell injury mechanism. Methods: HUVECs were divided into control group, fever group (F group), KD non-CALs group and KD combined coronary injury group (CALs group). Cell proliferation was detected by MTT assay. Apoptosis of HUVECs in each group was analyzed by flow cytometry (FCM). Results: The results of MTT assay showed that the cell viability of Non-CALs group and CALs group was lower than that of F group and Control group, and the CALs group was lower than Non-CALs group (P <0.01) . In addition, the apoptotic rate of HUVECs increased after serum of KD patients, which was significantly different from the control group (P <0.01). The apoptosis rate of CALs group was higher than that of Non-CALs group. Conclusion: Serum of KD patients with coronary artery injury can inhibit the proliferation of HUVECs and induce the apoptosis of HUVECs.
