以不同生长来源的3种“宁杞1号”枸杞苗为试材,通过比较其生长状态及特点,筛选诱导愈伤组织形成的外植体来源枸杞苗。同时,研究了不同激素组合对枸杞愈伤组织形成的影响。在此基础上,比较了以叶片和茎段为外植体,愈伤组织形成的时间及长势。结果表明:“宁杞1号”枸杞种子种植的无菌苗长势好,继代后性状稳定,可作为最佳外植体材料来源;以叶片为外植体,诱导愈伤组织形成时间短、长势好、不定芽形成早;初步确定诱导枸杞叶片形成愈伤组织的最佳培养基为MS+0.2mg·L~(-1) NAA+1.0mg·L~(-1) 6-BA。
Three kinds of “Ningqi No.1” wolfberry seedlings from different growth sources were used as materials to compare the growth status and characteristics of the Chinese wolfberry seedlings. The explants derived from callus were screened for Lycium barbarum. At the same time, the effects of different hormone combinations on the callus formation of Lycium barbarum were studied. On this basis, the time and growth of callus formation were compared by using leaves and stem segments as explants. The results showed that the sterile seedlings planted with Lycium barbarum “Ningqi 1” grew well, and the characters were stable after subculture, which could be used as the best explant source. The leaves were used as explants to induce callus formation time Short, good growth and adventitious bud formation. The best medium for inducing callus formation of Lycium barbarum was MS + 0.2 mg · L -1 NAA + 1.0 mg · L -1 6-BA .
