目的通过观察活性维生素D3对实验性自身免疫性肌炎小鼠是否有抑制作用,通过测量调节性T细胞(Treg)转录因子Foxp3和其相关的IL-10和TGF-β的mRNA表达,探讨其可能的作用及机制。方法将10只BALB/c小鼠随机分为模型组5只及活性维生素D3干预组(干预组)5只,均制备肌炎模型;干预组小鼠行活性维生素D3腹腔注射,同期模型组腹腔注射等体积生理盐水。在第十四天处死并取材,采用实时荧光定量PCR法检测二组脾脏组织中的Foxp3、IL-10和TGF-βmRNA表达。结果与模型组比较,干预组小鼠脾脏中的IL-10、TGF-βmRNA的表达增加(P<0.01);Foxp3 mRNA的表达无区别(P>0.05)。结论活性维生素D3可能通过增加Treg细胞相关因子IL-10和TGF-β的表达对肌炎小鼠发挥免疫保护作用。
Objective To observe whether the inhibitory effect of active vitamin D3 on mice with experimental autoimmune myositis was investigated by measuring the mRNA expression of Foxp3, a transcription factor of regulatory T cells (Treg), and its related IL-10 and TGF-β Possible role and mechanism. Methods Ten BALB / c mice were randomly divided into model group (n = 5) and active vitamin D3 intervention group (n = 5). Myocarditis models were prepared. The mice in the intervention group were injected intraperitoneally with active vitamin D3, Injection of equal volume of saline. Were sacrificed on the 14th day and drawn, and the expression of Foxp3, IL-10 and TGF-βmRNA in the two groups were detected by real-time fluorescence quantitative PCR. Results Compared with the model group, the expression of IL-10 and TGF-βmRNA in the spleen of mice in the intervention group increased (P <0.01), but there was no difference in the expression of Foxp3 mRNA (P> 0.05). Conclusion Active vitamin D3 may play an immunoprotective role in myositis mice by increasing the expression of IL-10 and TGF-β in Treg cells.
