为定量测定酶法合成龙胆低聚糖得率,采用葡萄糖氧化酶-过氧化物酶法(GOD-POD),通过测定转化后残留底物葡萄糖含量,间接测定龙胆低聚糖得率。对该法测定酶法合成龙胆低聚糖体系中残留底物葡萄糖含量的干扰因素、方法的稳定性、重复性、精密度进行研究。结果:该法在2h内测定体系葡萄糖含量结果稳定(RSD=0.81%),重复性好(RSD=0.97%),精密度高(RSD=1.55%),干扰小(加样回收率=101.681%)。结论:GOD-POD法用于测定酶法合成龙胆低聚糖体系中残留底物葡萄糖含量结果稳定可靠,体系中产物龙胆低聚糖及其他物质对GOD-POD法测定葡萄糖含量无显著影响。用该法间接测定酶法合成龙胆低聚糖得率可行。
In order to quantitatively determine the yield of gentian oligosaccharides, the glucose oxidase-peroxidase (GOD-POD) method was used to determine the yield of gentian oligosaccharides indirectly by measuring the glucose content of the substrate after transformation. The method was used to determine the interference factors of residual substrate glucose content in enzymatic synthesis of gentian oligosaccharide system. The stability, repeatability and precision of the method were studied. Results: The method was stable (RSD = 0.81%) with good repeatability (RSD = 0.97%), high precision (RSD = 1.55%) and small interference (sample recovery = 101.681% ). Conclusion: The results of GOD-POD method for the determination of residual substrate glucose content in enzymatic synthesis of gentian oligosaccharide system are stable and reliable. The gentiooligosaccharides and other substances in the system have no significant effect on the determination of glucose content by GOD-POD method . The method of indirect determination of enzymatic synthesis of gentian oligosaccharides yield is feasible.
