目的 探讨细胞间隙连接蛋白 Cx43在人子宫颈癌细胞系 He L a中表达 ,及其对细胞间隙连接通讯的影响 .方法 应用流式细胞仪以及 L ucifer Yellow划痕标记荧光传输技术 ,研究维甲酸作用对 He L a细胞 Cx43蛋白表达 ,以及细胞生长状态和间隙连接通讯功能的调节作用 .结果 He L a细胞经维甲酸处理后 ,流式细胞仪分析 ,Cx43蛋白荧光强度明显增强 ,Cx43蛋白表达阳性细胞计数率由 1.9%上升至2 6 .3% ;He L a细胞生长明显受抑 ;细胞间隙连接通讯功能得到恢复 .结论 维甲酸可通过上调 Cx43蛋白在 He L a细胞中的表达 ,使细胞间隙连接通讯功能得到恢复 ,实现对宫颈癌细胞恶性表型的逆转作用
Objective To investigate the expression of connexin Cx43 in human cervical carcinoma cell line HeLa and its effect on the gap junctional intercellular communication.METHODS Flow cytometry and L ucifer Yellow scratch-labeled fluorescent transmission technique were used to investigate the effect of retinoic acid On the expression of Cx43 protein and the regulation of cell growth status and gap junctional intercellular communication in HeLa cells.Results After treated with retinoic acid for HeLa cells, the fluorescence intensity of Cx43 protein and the expression of Cx43 protein were significantly increased by flow cytometry The positive rate of positive cells increased from 1.9% to 26.3%, the growth of HeLa cells was inhibited obviously, and the communication function of cell gap junction was restored.Conclusion ATRA can up-regulate the expression of Cx43 protein in HeLa cells, Cell gap junctional communication function was restored, to achieve the reversal of malignant phenotype of cervical cancer cells
