目的:建立并比较研究不同药用部位茯苓中三萜类成分的UPLC-UV-MS指纹图谱。方法:以去氢茯苓酸为对照品,采用Shim-pack XR-ODSⅡ(75 mm×2.0 mm,2.2μm)色谱柱,以乙腈-0.1%甲酸为流动相梯度洗脱;检测波长242 nm,流速为0.5 mL/min,柱温30℃;采用SPSS 16.0进行聚类分析。结果:建立茯苓中三萜类成分的UPLC-UV-MS指纹图谱,共标定22个峰,其中12个为共有峰,方法学考察结果符合指纹图谱技术要求;相似度和聚类分析结果表明,茯苓皮和茯苓块中三萜类成分存在显著差异,茯苓块药材质量与其产地有关,而茯苓皮药材不同产地间差别不大。结论:该研究方法稳定、可靠、重现性好,可用来控制茯苓的质量。
Objective: To establish and compare UPLC-UV-MS fingerprints of triterpenoids in Poria cocos with different medicinal parts. Methods: Hydrogenpolysaccharidide was used as a reference substance and was eluted with a mobile phase of Shime-pack XR-ODSⅡ (75 mm × 2.0 mm, 2.2 μm) using acetonitrile-0.1% formic acid as mobile phase. The detection wavelength was 242 nm and the flow rate 0.5 mL / min, column temperature 30 ℃; using SPSS 16.0 cluster analysis. Results: UPLC-UV-MS fingerprints of triterpenoids in Poria cocos were established. Twenty-two peaks were identified, of which 12 were common peaks. The results of the methodological studies were in accordance with the fingerprinting requirements. The results of similarity and cluster analysis showed that, Tuckahoe and Poria block triterpenoids significant differences, the quality of Fuling block medicinal herbs and its origin, and Poria skin medicinal materials between different origin is not different. Conclusion: The research method is stable, reliable and reproducible. It can be used to control the quality of Poria.
